Abstract

Background: Osteoarthritis (OA) has been recognized, as the most common inflammatory disease in the world. Adipose Stem Cells (ASCs), as a new feasible source with high numbers of stem cells and proliferative capacity have been used for regenerative medicine. Based immunomodulatory and chondrogenic properties of ASCs, this study aimed to assess intra articular injection of ASC effect on improvement of osteoarthritis signs. Methods and Materials: Adipose tissue samples were obtained from subcutaneous of abdomen. ASCs were isolated and cultured for at least three passages in culture media containing autologous serum and expanded them to 15-20 × 106 cell. The morphology and proliferative potency of ASCs were determined. Immuno phenotype characteristics of ASCs were analyzed by flow-cytometry. Then cell suspensions were injected into knee articular spaces. After 6 months the function of knee was assessed by WOMAC, KOOS, Lysholm and Lequesne indexes. Results: The results of this study showed that homogenous spindle-shape ASCs expanded rapidly with low doubling time. The low expression of CD14 and CD45 indicated that ASCs are non hematopoietic cells and expressed high percentages of CD44, CD105 and CD90. Our results showed that injected ASCs were effective in improvement of OA by scoring systems for evaluation of pain, joint movements and daily physical activities were significantly changed due to injection of stem cells. Osteoarthritis severity indexes means of WOMAC and Lequesne were decreased from 53 to 12.3 and 15.1 to 2.1 respectively. Also osteoarthritis improvement indexes Lysholm & KOOS means were significantly increased from 35 to 15.1 and 70 to 126.7 respectively. In six months follow up of intra articular injection of ASCs, we observed no local or systemic side effect. After ASCs injection, walking distance considerably increased. The flexion angle of knee improved by 20-30 degrees compares to before of treatment. Conclusion: Autologous ASCs injection could be resulted in increasing of knee function, alleviated of pain and quality of life improvement.

Abstract

The presence of melamine in foods is one of the important issues in terms of food safety and consumer health. Extraction is the first step in determining the amount of melamine in foods. Therefore, in the current study, melamine extraction from egg was optimized using response surface methodology based on the central composite design. The percentage of water used in the extraction phase containing water/acetonitrile, extraction phase volume, extraction time, pH of extraction phase, and hydrochloric acid concentration in the extraction phase were considered as the studied variables for optimizing extraction conditions. Finally, the amount of melamine present in four brands of egg was measured by high-performance liquid chromatography (HPLC) using internal standard method. Optimum extraction conditions were obtained in the form of 39.28% water in the extraction phase, 5.06 mL extraction solvent volume, 12.48 min extraction time, pH 5.04 and acid concentration of 0.73 M. The amount of melamine in the egg samples was in the range of 0.19-0.45 mg/kg. Regarding the amount of melamine authorized by the EU and the US Food and Drug Administration, it can be concluded that the melamine content of the samples is within the limits of the EU standard and only in two samples was more of the USFDA standard.

Abstract

Water deficit is a major factor limiting plant productivity of fruit quantity and quality, particularly in arid and semi-arid regions of the world. The total yield, fruit physical characteristics and nutritional value of four drought tolerant tomato genotypes (KSU-TOM-102, KSU-TOM-106, KSU-TOM-107 and TL–01860) were evaluated in response to deficit irrigation (DI) under field conditions. Three levels of crop evapo-transpiration (ETc; 50%, 75% and 100%) were applied at three different growth stages (vegetative, flowering and fruiting stage) to the four tomato genotypes. Tomato genotypes differed in their responses to water deficit. Among different genotypes, KSU-TOM-102 recorded the highest average total yield (89.54 t.ha-1) under irrigation treatment with 100% of ETc during all stages as well as 75% of ETc during the fruiting stage. But, in general, total yield decreased under water deficit. Significant (P≤0.05) differences in fruits quality characteristics exerted by irrigation water treatments, tomato genotypes and their interactions. Irrigation with water at 50% ETc at all growth stages significantly (P≤0.05)  increased vitamin C, titratable acidity, total soluble solids and total sugar contents for tomato ‘KSU-TOM-107’ followed by ‘KSU-TOM-102’. But, this increase in nutritional value was accompanied with decrease in total yields by nearly 40-50%.   KSU-TOM-102 irrigated with water at 75% ETc at fruiting stage or vegetative stage recorded 0% and 12% reduction of the total yield, respectively, while maintaining good nutritional value as compared with 100% of ETc during all stages. Therefore, these treatments can be recommended as irrigation management strategy for tomato ‘KSU-TOM-102’ production under field conditions.
