Abstract

Background and aims: The E2F family of transcription factors is encoded by at least eight genes, E2F1-8. These proteins are important targets of the retinoblastoma protein (RB) contributed in regulation of transcription, cell cycle and apoptosis. The aim of this study was to investigate the expression levels of E2F protein family including E2F1, E2F2, E2F7, E2F8 and RB1 in the lineage negative (Lin-) hematopoietic stem cells (HSCs) of young and aged black mice using Real Time RT-PCR and western blot techniques. Methods: Lin- HSCs of 4 young (7-10 weeks old) and 3 aged (76 weeks old ) mice have been isolated from their bone marrow cells using MACS column and after RNA extraction of culturing cells and cDNA preparation, samples were then analyzed by Real Time RT- PCR and western blot techniques. Results: The E2F7 and E2F8 expression levels of the Lin- HSCs of old mice were only the transcriptional factors significantly decreased when compared with young mice. In conclusion: It seems the functional roles of important E2F7 and E2F8 transcription factors in moderating potentially destructive activity of E2F1 and regulation of cell cycle have been diminished in Lin- HSCs of aged mice. Hence, the apoptotic activity of the E2F1 would affect to the most HSCs, reinforcing bone marrow to proliferate the HSCs in old mice. However, Real Time RT-PCR data showed that the expression level of the E2F1 in those cells was not increased significantly as expected. This is the first report in this regard and further investigation with more samples need to reconfirm these data.

Abstract

 The aim of this study was to determine Total Phenolic Compound (TPC) and antioxidant activity changes depending on the ripening in Rosa canina, R. dumalis, R. dumalis ssp. boissieri and R. villosa rosehip species. Rosehip fruits were harvested in 6 different times from July to September. TPC content and antioxidant activities were determined by spectrophotometric methods. TPC and antioxidant activities of the studied species increased during ripening. TPC (1510.57 mg GAE 100 g-1/H-6) and antioxidant activities (TEAC: 364.12 µmol trolox equivalent g-1/H-6 and FRAP: 286.79 µmol trolox equivalent g-1/H-6) were higher in R. dumalis (MR-15) than the other studied species. There was a high correlation between the TPC and the antioxidant activity. Also, there was a positive correlation between maturation, phenolics and antioxidant activities. This correlation was high in R. dumalis and low in R. canina. However, it was not possible to express the existence of a relationship between temperature, TPC, and antioxidant activity. It was possible to express the existence of correlations between the color of fruits and some studied characteristics. The correlations between the colors of the fruits and TPC and antioxidant activity of R. dumalis were found higher than the other species.

Abstract

This study was conducted to determine the changes in total sugar, ascorbic acid, α-tocopherol and β-carotene content of fresh fruits of rosehip species during ripening, for three years. Fruits of selected genotypes belonging to Rosa dumalis, R.canina and R.villosa were used. Fruits were harvested in six different times based on hyphantium color change or flesh softening. The total sugars of R.dumalis, R.canina, and R.villosa fruits ​​ranged between 11.22-16.68, 9.28-13.90, and 9.28-16.31%, respectively. Also, ascorbic acids varied between 767.1-1324.9, 168.7-481.5 and 241.2-574.2 mg 100 g-1, respectively; α-tocopherols ranged between 5.87-10.25, 5.29-10.04 and 2.74-11.65 mg 100 g-1, and β-carotenes varied between 0.18-2.03, 0.34-2.42 and 0.49-3.62 mg 100 g-1, respectively. Total sugar, ascorbic acid, α-tocopherol and β-carotene content linearly increased depending on ripening time. Relationships between total sugar and vitamins were significant. It was concluded that harvest should be delayed as much as possible in order to obtain fruits with high content of total sugar and the main components of vitamins.
